EMT6

  • a transplantable mouse mammary carcinoma that arose in a BALB/cCRGL mouse after implantation of a hyperplastic mammary alveolar nodule
  • maintained in MEM/F12 + 10% FBS
  • passaged by mild treatment with 0.25X trypsin/EDTA and diluting into fresh media (1:10) every 3 to 4 days

MCF7

  • HTB-22
  • mammary adenocarcinoma
  • MEM + Earle's Salts + 10% FBS
  • Trypsin. Quench with media. Transfer the cell suspension to the centrifuge tube with the medium and cells, and centrifuge for 5 minutes. Resuspend the cell pellet in fresh growth medium. Add appropriate aliquots of the cell suspension to new culture vessels.

MCF10A

  • CRL-10317
  • non-tumorigenic epithelial cell line
  • Differentiation protocol
  • DMEM/F12 + 2.5% Horse serum + 0.5ug/mL hydrocortisone + 10ug/mL Insulin + 20ng/mL EGF + 100ng/mL Cholera toxin
  • Trypsin at 37C. Quench with media. Transfer the cell suspension to the centrifuge tube with the medium and cells, and centrifuge for 5 minutes. Resuspend the cell pellet in fresh growth medium. Add appropriate aliquots of the cell suspension to new culture vessels.

MDA-MB-231

Cervix

HeLa

  • cervical carcinoma

    • CCL-2 is directly from ATCC, adherent
    • SEN2 is kindly provided by DiMaio lab, adherent
    • S3 from Maniatis Lab, suspension
  • CCL-2 and SEN2 are maintained in DMEM + 10% FBS and passaged by mild treatment with 0.25X trypsin/EDTA and diluting into fresh media (upto 20 fold dilution is OK)
  • S3 cells are maintained in RPMI + 10% FBS and passaged by dilution (1:10)


Colon

CT26.WT

  • CRL-2638
  • an N-nitroso-N-methylurethane-(NNMU) induced, undifferentiated mouse colon carcinoma cell line
  • RPMI-1640 + 10% fetal bovine serum (55mL for 555mL)
  • passaged by mild treatment with 0.25X trypsin/EDTA and diluting into fresh media (1:10) every 3 to 4 days

HCT116

  • CCL-247
  • colorectal carcinoma
  • maintained in McCoys 5A + 10% FBS
  • passaged by mild treatment with 0.25X trypsin/EDTA and diluting into fresh media (1:8) every 3 to 4 days
  • freeze medium: complete growth medium supplemented with 5% (v/v) DMSO

HT29

  • colorectal carcinoma
  • three lines available: HT29, HT29-bGAL, HT29-APC
  • HT29-bGAL and HT29-APC are provided by Bert Vogelstein are described in Morin et al
    • HT29-bGAL and -APC lines contain zinc inducible (metallothionein promoter) beta-galactosidase or adenomatopolyposis coli transgenes, respectively
    • maintained in McCoys 5A media + 10% FBS + 0.6mg/mL Hygromycin (Invitrogen)
  • HT29 is the parental line
    • maintained in McCoys 5A media + 10% FBS
  • passaged by mild treatment with 0.25X trypsin/EDTA and diluting into fresh media (1:8) every 3 to 4 days
  • avoid prolonged passage
