Primary ChIP is performed as described. ChIP DNA elution step is modified and does not use SDS.
- Add 100uL 20mM DTT in TE to washed beads
- Incubate 30 minutes at 37°C with mild mixing
- Transfer the elution to new tube and dilute the elution in 1mL IP buffer (may also remove DTT using desalting columns)
- Add detergent, DOC and protease inhibitors, as in the first immunoprecipitation step
- Perform the second ChIP
